Karta!Föredragshållare: Lukas Käll, Center for Biomembrane Research, Dep. of Biochemistry and Biophysics Stockholms Universitet Titel: A semi-supervised method to assess and improving confidence in identifications in shotgun proteomics Abstract Shotgun proteomics uses liquid chromatography-tandem mass spectrometry to identify proteins in complex biological samples. Here we first describe an algorithm, called Percolator, for improving the rate of confident peptide identifications from a collection of tandem mass spectra. Percolator uses semi-supervised machine learning to discriminate between correct and decoy spectrum identifications. We will describe a recent extension of the feature set of percolator, were we use the deviance between predicted and observed retention time of the peptides as an indicator of correctness. |
